NG2 and DLL3 CAR-T Cells Targeting Melanoma

Safety and Efficacy of NG2 and DLL3 CAR-T Therapy Targeting Melanoma

NG2 and DLL3 CAR-T Cells Targeting Melanoma (NCT07193966) is a Phase 1 / Phase 2 interventional studying Melanoma, sponsored by Shenzhen Geno-Immune Medical Institute. RECRUITING as of the most recent ClinicalTrials.gov update. Talk to your doctor before contacting the trial site.

About This Trial

The purpose of this study is to assess the feasibility, safety and efficacy of multiple CAR-T cell therapy which targets NG2 and DLL3 surface antigens in patients with relapsed and refractory melanoma.

What Stage of Research Is This?

Phase 1 trials test a new treatment for the first time in humans, focusing on safety, dosing, and how the body processes the drug. For Melanoma, a Phase 1 study typically enrolls a small number of participants — often healthy volunteers or patients who have exhausted standard treatment options. Phase 1 results determine whether a treatment moves into larger Phase 2 efficacy studies.

This trial is currently recruiting participants. The sponsor has registered the study with ClinicalTrials.gov as actively enrolling, which means new applicants who meet the eligibility criteria can be considered for screening. Trial status can change between updates — confirm current recruiting status with the study contact before traveling for a screening visit.

Target enrollment of 100 participants puts this in the typical range for a Phase 2-style efficacy study or a moderate Phase 3 trial in a focused Melanoma subpopulation. At this scale, the study has enough statistical power to detect a clear treatment effect but is not the largest cohort in the field.

Who May Be Eligible (Plain English)

Who May Qualify: - Patients with tumors have received standard first-line therapy and have been diagnosed with non-resectable, metastatic, progressive or recurrent conditions. - The expression status of NG2 and DLL3 antigens of the tumor has been determined for eligibility. Positive expression is defined by NG2 and DLL3 antibody staining results based on immunohistochemistry or flow cytometry analyses. - Body weight greater than or equal to 40 kg. - Age: ≥18 year and ≤ 75 years of age at the time of enrollment. - Life expectancy: at least 8 weeks. - Prior Therapy: 1. There is no limit to the number of prior treatment regimens. Any grade 3 or 4 non-hematologic toxicity of any previous therapy must have resolved to grade 2 or less. 2. Participants must not have received hematopoietic growth factors for at least 1 week prior to mononuclear cells collection. 3. At least 7 days must have elapsed since the completion of therapy with any biologic agent, targeted agent, tyrosine kinase inhibitor or metronomic non-myelosuppressive regimen. 4. At least 4 weeks must have elapsed since prior therapy that includes a monoclonal antibody. 5. At least 1 week must has elapsed since any radiation therapy at the time of study entry. - Karnofsky/jansky score of 70% or greater. - Cardiac function: Left ventricular ejection fraction greater than or equal to 40/55 percent. - Pulse Ox greater than or equal to 90% on room air. - Liver function: defined as alanine transaminase (ALT) \<3x upper limit of normal (ULN), aspartate aminotransferase (AST) \<3x ULN; serum bilirubin and alkaline phosphatase \<2x ULN. - Renal function: Patients must have serum creatinine less than 3 times ULN. - Marrow function: White blood cell count ≥1000/ul, Absolute neutrophil count ≥500/ul, Absolute lymphocyte count ≥500/ul, Platelet count ≥25,000/ul (not achieved by transfusion). ...See full criteria on ClinicalTrials.gov Always talk to your doctor about whether this trial is right for you.

How to Talk to Your Doctor About This Trial

Bring the printable summary of this trial — including the NCT ID (NCT07193966), the sponsor (Shenzhen Geno-Immune Medical Institute), and the key eligibility criteria — to your next appointment. Your doctor can review the inclusion and exclusion criteria against your medical history, lab values, and current treatments to assess whether you are likely to qualify. They can also help you weigh whether trial participation makes sense alongside your existing care plan.

Useful questions to walk through together: What does the trial protocol require beyond standard care? How long is the active treatment phase, and how long is follow-up? Are there study visits at sites I can reach? Who pays for the trial-specific procedures, and who pays for standard-of-care portions?
